CircRNA RT-PCR Detection Kit

Intended Use

CircRNA RT-PCR Detection Kit is an in virto diagnosis device intended for quantitative detection of four CircRNA gene in human saliva sample. The test result act as an aid in the diagnosis of depression, the CircRNA RT-PCR Detection Kit should not be the only basis to diagnose or exclude depression, and can not be used for screening.

For professional use only, The kit should be use in qualified PCR laboratory.

Test Principle

This kit is based on real-time fluorescence quantitative PCR technology, Specific primers are designed according to the highly conserved regions and cyclization sites of four circRNA biomarkers to amplify the corresponding nucleic acid fragments. At the same time, highly specific TaqMan probes are used to bind to the corresponding nucleic acid fragments, and hydrolysis occurs under the action of Taq enzyme exonuclease activity, generating fluorescent signals. Real time amplification curves are obtained based on the relationship between fluorescent signals and amplification cycles, achieving the detection of the content of the four circRNA biomarkers in the sample.

The PCR detection system of this kit includes endogenous housekeeping gene internal standard (IC) primers, which can monitor specimen sampling, RNA extraction, and detection processes by detecting whether the internal standard is normal, avoiding false negative results.

Applied Instruments

This kit can be use on PCR instruments contain the detection channel of FAM, ROX and CY5. For example, ABI 7500, HONGSHI SLAN-96S.etc

Storage and shelf life

1. The kit can be stable for 12 month when storage avoid light under -20±5℃

2. Please refer to the outer package label for the production and expiration date.

3. The kit should not be freeze and thaw for more than 3 times.

4. Please transfer the kit with ice in a curling bottle or sealed with ice in a foam box.

2. Sampling method

Rinse your mouth with water 30 minutes before collection, it is strictly forbidden to collect immediately after rinsing. use your tongue to press against the root of your upper or lower teeth to enrich saliva, and gently spit saliva into the oval funnel until the liquid saliva (non bubble) reaches the filling line height of 2 mL. Carefully unscrew the funnel from the collection tube, Please always keep the collection tube upright during operation. Tighten the red cap and seal the collection tube. Check whether the sample collection tube is intact, whether there are any cracked tubes, open lids, or other sample spillage situations. Discard the used funnel.

3. Sample storage

The sample collected should be used for test immediately.

If not, The sample can be stable under 2~8 ℃ for 72 h, for 3 months under -20 ℃, or for 12 months under -70 ℃.

Please do not freeze-and-thaw the sample for more than 3 times.

4. Sample transferring

Please transfer the kit with ice in a curling bottle or sealed with ice in a foam box.
